The transdermal delivery system(TDS) of traditional Chinese medicine (TCM) is made of the effective component extract with compatible subsidiary material in the guidance of TCM theoretics. The effective component of the TCM penetrate from the skin surface and get into each layer of the skin in a steady speed and then enter the blood circulation to form the local or systemic therapy function by administering the TDS on the skin. The new Formulations of TDS used for TCM such as patch, gel, cataplasma etc, they are convenient and safe with reliably curative effect and have been used in clinic.The aim of the research is preparation of the new hot melt press sensitive adhesive which used in the TCM TDS. First, the effect of the main component on the adhesion property of SIS hot melt press sensitive adhesive (HMPSA) was studied to select the optimal prescription. Then studied the effect of a lipophilic plasticizer DOP on the softening point and microstructure, and then studied the adhesion and drug release properties of SIS HMPSA. In the end, on the basis of relatively low softening point and favorable adhesion property of SIS HMPSA, an acrylic resin RLPO was blended with it to form the bi-continuous structure. The effect of constitute, proportion and preparation technics on the microstructure, adhesion property and softening point was investigated respectively, and the drug release from the bi-continuous HMPSA was also researched.The results showed that when the SIS:C5:mineral oil=3:5:2, the HMPSA had the best performance. The addition of DOP could decrease the softening point of HMPSA. When the amount of DOP was up to 20%wt of SIS the softening point of the HMPSA decreased by 17℃and kept appropriate adhesion property. The drugs with high logP, low molecular weight and melting point were advantaged for their release from the HMPSA. DOP improved the release of the lipophilic drug, but inhibited the release of the hydrophilic drug. When the proportion of the SIS/RLPO was 1/1 with SMA up to 2.5%wt, the bi-continuous formed by blending, and provided the material with favorable adhesion property. The bi-continuous structure was advantaged for the release together of effective component with different polarity in the TCM.
