Preparation Of Xiaoshipingwei Granul And Its Effect On Growth Of Puppies

Xiaoshipingwei Powder, included in the 2005 edition of The People's Republic of China Veterinary Pharmacopoeia", composed by Semen Arecae, Fructus Crataegi, Rhizoma Atractylodis, Pericarpium Citri Reticulatae, Cortex Magnoliae Officinalis, Radix Glycyrrhizaei, can increase appetite, promote gastrointestinal peristalsis, and enhance the immunity. In order to promote the digesting effects and product quality control standards, this proved recipe is changed from the powder into granules, and named Xiaoshipingwei Granule. According to higher essential oil content and granule preparation characteristics, Xiaoshipingwei Granule was prepared through the volatile oils extraction andβ-CD inclusion, the active ingredient content was determinated by UV spectrophotometry, and the acute toxicity of Xiaoshipingwei Granule and its effect on the growth of puppies also were explored.Through the orthogonal test, we studied the best extracting process of the volatile oil in Rhizoma Atractylodis, Pericarpium Citri Reticulatae by steam distillation, the best extraction process was for medicinal powder 40 mesh sieve, adding 10 times the amount of water, soaking 0.5h, extracting 5h. Ultrasonic method was used for volatile oil B-CD inclusion, and the best inclusion condition was oil:β-cyclodextrin 1:8, inclusion temperature 40℃, Ultrasonic time 60min. Percolation method was used for the ingredient extracting of betel nut etc., and the best extraction process was 60% ethanol and ethanol as 8 times the amount of herbs, soaked 12 hours, the flow rate 3ml/min.The content of arecoline was determined in the extracts by UV spectrophotometry, the result showed that the higher alkali content using percolation method for extraction of Semen Arecae and other ingredients. The screening tests for preparation method of Xiaoshipingwei Granule was made for determining of ratio of particles of raw materials, alcohol concentration, alcohol consumption for preparation process. The preparation process was raw materials:Dextrin:sucrose as 1:0.5:1,70% alcohol concentration, alcohol the total amount of raw materials accounted for 16%. The resulting product is a good particle shape, particle size uniformity, high yield, can meet the production needs.The appearance, particle size, moisture content, melting resistance of Xiaoshipingwei Granule was tested, the target particles reached the Veterinary Pharmacopoeia of Chinese medicine granules on the relevant requirements, consistent with its color, grain uniform, no moisture absorption caking and other phenomena. Determination of alkali content in Xiaoshipingwei Granule was established by UV spectrophotometry, To the absorbance for the vertical and density for horizontal, standard curve regression equation:A= 3.4936C+0.0301, R2= 0.996, results showed the good linear relationship in the concentration 0.0472-0.236mg/mL. The detection method with good reproducibility, high precision, can be used as the standard content determination of Semen Arecae.Acute toxicity experiments showed that Xiaoshipingwei Granule had no side effect, no any anatomical changes for testing mouse. The effect of Xiaoshipingwei Granule on the growth performance of puppies was tested in comparasion with Xiaoshipingwei Powder group and control group. The result showed that Xiaoshipingwei Granule icreased weight gain 16.77%, feed conversion rate 12.92% compared with Xiaoshipingwei Powder group, and weight gain 25.76%, feed conversion rate 25.28% compared with contol group.
