Optimization of Bacillus licheniformis L3 fermentation medium in this research. Based on the single-factor test, carbon source, soybean powder and K2HPO4 were determine as necessary for spore formation of Bacillus Hcheniformis L3 through the Plackett-Burman design. The medium consist with soybean flour 8g/L, K2HPO4 2g / L, carbon 32.20g/L, MgSO4 0.5g/L, yeast extract 2.5g/L were defined as the optimum medium for Bacillus Hcheniformis L3 growth and spore formation, through the center combination of design and response surface analysis. Fermentation with the optimummedium achieved live bacteria 100.2×108 CFU/mL, nearly double than that with the initial culture medium.Through optimization of culture conditions, the best inoculum of liquid was 4%, 25mL/250mL, speed on 200r/min, use the defoamer oil (from 1000 mg/L to 3000 mg / L) or the bubble enemy (from 100 mg/L to 300 mg/L)to elimination the bubble. On the 16L fermentor, the number of viable cells 92.7×108 CFU/mL, increased 70.09% than the initial(52.7×108 CFU/mL) one.Preliminary experiment showed that the Bacillus Hcheniformis could excite the growth of some crops. This topics carried out the research on the yield increase on corn,cotton, rice and cabbage by the fermentation broth, supernatant and pellet of Bacillus Hcheniformis L3 respectively. Results showed that the fermentation broth of Bacillus Hcheniformis L3 significantly increased the production of corn,cotton etc.. Furthermore, This is mainly caused by the pellet of the fermentation broth of Bacillus Hcheniformis L3. The effect of upland crops were better than paddy field crops.
