The best process of extracting protein from rice residue with enzyme was studied.The obtained protein powder,on the one hand,was used to prepare high emulsibility production by glucosylation modification and,on the other hand,was used as part of wall materials,The optimal formula and Process Parameters of microcapsule powdered oil with high rice protein content were researched,and the prepared product meets the qualification of powdered milk for feeding.1.The physic-chemical indexes of rice residue were determined.The results showed that protein,carbohydrate and fats were its main compositions.The optimum conditions of extracting rice residue oil with Hexane were temperature 20℃, material-liquid ratio 1:7(W/V),extraction time 2h.Under these conditions,the extraction rate could reach 8.85%.The fatty acid composition of rice residue oil was analyzed:saturated fatty acids 25.5%,unsaturated fatty acids 73%.Oleic acid occupyed the highest content of unsaturated fatty acids,the ratio was 36.4%.The percentage of oleic acid and linoleic acid was 1:1.04,which accorded with the optimum proportion 1:1 that WHO recommends.2.Degreasing rice residue was washed by hot water and then filtered,the determination method of sugar content(anthrone-sulfuric acid method) in filter liquor was tested by stability,recovery,reproducibility and the results indicated that anthrone-sulfuric acid method fitted the demands of analysis,Optimization of operating conditions was performed in the extraction of water-soluble sugar from rice residue.The results were as follows:the material-liquid ratio was1:8,temperature 80℃,extraction time 25min and eluting times n=2.3.To study the effects of different enzyme dosage,reaction temperature,pH and reaction time in the extraction of protein from rice residue which was disposed by degreasing and washing,The experience of extracting protein by enzyme was carried out.Response surface methodology was used to optimize the hydrolytic conditions, which for Alcalase 2.4L FG were 61.43℃,pH8.47,enzyme dosage 1.85%, substrate/solvent ratio of 10%(W/V),reaction time of 2.82h.Within these conditions, the protein content(dry basis)could reach 90.1%,The solubility of protein was 90.3% and DH could achieve 16%.The property of rice residue protein was fine,but the sense-organs and taste were not satisfied.4.Debittering and decolorant tests of rice residue protein power by enzymatic hydrolysis were carried out.Debittering tests showed that bitterness could basically removed when the amount of flavourzyme was no less than 0.4%,and the product had a certain fresh flavor;The best destained conditions of the product were pH10, H2O2/water ratio of 0.06%(V/V),temperature 40℃,decolorant time 2h.Amino acids in destaining solution and hydrolysate were determined,It could be told from the results that both of them had the same kinds of amino acid,and the contents were nearly the same.It indicated that H2O2 had no destructive effect during the process of decoloration.Little changes of the content pre and post decoloration in hydrolyzed protein powder showed that decolorization method was feasible.5.Glycation reaction was made between DH 16%enzymatic hydrolyzed rice residue protein and lactose.On the basis of single factor experiment,the orthogonal trial was used in optimization of technological parameters.The optimum technological conditions were as follows:the mass ratio of enzymatic hydrolyzed rice residue protein to lactose 3:1,reaction temperature 80℃,pH 8,reaction time 8d.6.The preparation of micro encapsulation powdered oil was conducted with sodium caseinate as the main emulsifier and enzymatic hydrolyzed rice residue protein power,lactose and maltodextrin as wall materials.The optimum recipe for emulsifier was studied:sodium caseinate:monoglyceride:modified soya lecithin was 15:6:4;The formula for powder oil products was as follows:fats 26%,lactose 28%, maltodextrin(dry basis)11%,hydrolyzed rice residue protein power 30%,sodium caseinate 3%,modified soya lecithin 1.2%,monoglyceride 0.8%.The appearance, physicochemical index and restoration emulsion were highly similar to milk powder and imported milk replacer of early weaning piglets(import price:20000 Yuan/t).It has significant market prospects and economic benefits on the diet of early weaning piglets.
