Objective: The aim of this work was to prepare HGF loaded polylactic acid and oxygen carboxymethylated chitosan copolyer (PLA-O-CMC) nanoparticles, to characterize them, to explore their degradation in vitro release, and we studied the effect of the nanoparticles on the viability and function of cultured rat hepatocytes.Methods: HGF loaded PLA-O-CMC Nanoparticles were prepared by ultrasound combine magnetic stirring method. The nanoparticles were characterized by electron microscopy, Laser Particle Size Analyzer and Zeta Potential Analyzer. TEM was used to study the morphological change of the NPs during the degradation, the weight loss of NPs and pH change of degradation wedium were also observed as well. The vital force and function of the cultured rat hepatocytes were observed by Cell Counting Kit-8 and biochemistry analysator every 24 hours within 1 week.Results: The results of characterizing HGF loaded PLA/O-CMC NPs showed that the mean size of NPs was 139.82nm, polydispersity was 0.108, HGF-loading level was 0.12665%, HGF-embedding ratio was 76.32%, and the zeta potential was 32.8eV. The investigation of HGF release behavior from NPs showed that: (1) the release kinetic equation of HGF from the NPs in early 24 hours was Q=148.4266+189.0493 t1/2 ( R=0.97589 ) and coincided with Huguhui release equation; (2) the release kinetic equation of HGF from the NPs in 30 days was Q=1086.28966+58.23938t(R=0.99716)and coincided with zero-level release equation. The viability and function of rat hepatocytes cultured with HGF loaded PLA-O-CMC NPs are higherly than control groups.Conclusion: HGF loaded PLA-O-CMC NPs were successfully prepared, and the HGF loaded PLA-O-CMC NPs can effectively maintain the vitality and function of cultured rat hepatocytes.
