| In this paper,taking the sludges of Fumin hoggery in the Jingbei direct and the sludges of fertilizer plant of Chongming County, Shanghai, Three strains which were good at degrading ammonia was isolated after taming and enrichment. Based on analysis of phenotype, physiological and biochemical characters and 16SrDNA, Three strains was identified. Then the degrading characters of ammonia were studied. Construct of compound bacteria by these strains and the degrading characters of ammonia were studied systematically. The results could be summarized as follows:The microorganisms were isolated which could remove ammonia efficiently by means of enrichment culture technique. After secondary screening, three strains CM3,CM8 and WSC5 which could remove ammonia more efficiently were obtained. Among them, strain CM3 and strain CM8 were isolated from the sludges of Fumin hoggery; strain WSC5 was isolated from the sludges of fertilizer plant.Based on analysis of phenotype, physiological and biochemical characters and 16S rDNA,CM3 was identified as Bacillus amyloliquefaciens, CM8 was identified as Bacillus licheniformis, and WSC5 was identified as Bacillus subtilis.The effects of degradation time, temperature , pH,initial concentration of ammonia, inoculation amount ,carbon source and ratio of carbon to nitrogen on degradation was determined. The results showed that the initial concentration of ammonia,pH, carbon source and ratio of carbon to nitrogen have the important influence on the degradation rate of strains.The degradation rate of strain CM3 reached up to 45% in 4 days under the conditions of initial concentration of ammonia 400mg/L,pH7.5, glucose and sodium succinate as carbon source with ratio of 1:1, ratio of carbon to nitrogen 8:1. The degradation rate of strain CM8 was very close to 50% in 4 days under the conditions of initial concentration of ammonia 400mg/L,pH7.5, glucose and sodium succinate as carbon source with ratio of 1:1, ratio of carbon to nitrogen 8:1. The degradation rate of strain WSC5 was very close to 50% under the conditions of low initial concentration of ammonia,for instance the initial concentration of ammonia was 100mg/LThe degradation rate of strain WSC5 was very close to 40% in 4 days under the conditions of initial concentration of ammonia 400mg/L, pH8.0, glucose and sodium succinate as carbon source with ratio of 1:1, ratio of carbon to nitrogen 8:1.The three strains were mixed in different proportion to construct the compound bacteria.The degradation conditions of the compound bacteria showed that the degradation rate was improved compared with single strain. The compound bacteria which three strains were mixed in proportion 1:1:1 have efficient degradation rate. The degradation rate of compound bacteria reached up to 60% under the conditions of initial concentration of ammonia 600mg/L, pH7.5 and temperature 30℃. |
