The soybean dregs were used as raw materials in this paper,the alcohol precipitation method and membrane separate technology were employed to extract,separate and purify Soluble Soybean Polysaccharides(SSPS) from soybean dregs respectively.As a relult,the yield of SSPS extracted by alcohol precipitation method was up to 28.8%,while the yield of SSPS extracted by membrane separation tech was up to 23.6%.Meanwhile,the microstructure analysis and physical properties of SSPS were studied as well.AFS-930 atomic fluorescent photometer,AA2610 Atomic Absorption Spectrometer and auto hydrargyrum analyzer were used to assay the content of Hg,Pb,Cd,As in SSPS,the content was 0.0002mg,0.0050mg,0.0700mg and 0mg respectively.SSPS were separated and purified by DEAE-cellulose column chromatography.The results showed SSPS were composed by several polysaccharides.Average molecular weights filtrated by different membrane were 271.9kDa,39.3kDa,2.5kDa respectively analyzed by gel-permeation chromatograph(GPC),classification effect of SSPS was distinct;Fourier transformation infrared spectrum analysis(FT-IR) was used,results showed that atoms of SSPS were connected by different bands and functional group variety was also confirmed.Atomic Force Microscopy(AFM) was used to observed stereo-microstructure of SSPS.Results showed that the shape of SSPS molecular was circularity or hollow ball.Research ablout physical property showed that,SSPS have good emulsion activity(EA) and emulsion stability(ES);also could improve the stability of protein.The Granularity distributions of SSPS detected by Microtrac Nanotrac-150 Granularity determinator were 1574nm and 475nm, 41.6%and 58.4%respectively.Fermentation experiments of lactobacillus showed,SSPS were beneficial for lactobacillus about proliferation effect,metabolism activity and acid producing ability.The above investigation shows that SSPS can be separated and molecular-weight gradated by membrane separation technology efficiently.The products' structure and performance are stable.
