This paper studied preparation technology of biochemical reagent-peptone by enzynolysis fish surimi,which was picked out in the process of chilled Tilapia slices. Enzymatic hydrolysis conditions of AS.1398 neutral protease, papain, protamex, flavourzyme and trypsin were studied. The research contents and results in details showed as follows:1. Single factor tests were adopted to obtain conditonal parameters and optimun scope for application.Results showed that AS.1398 neutral protease, papain, protamex were rather appropriate.Optimization hydrolysis craft condition of AS.1398 neutral protease was as following: temperature was 55℃, pH value was 7.5, time of hydrolysis was 4h, input amout of enzyme was 750U/g. Optimization hydrolysis craft condition of papain was as following: temperature was 65℃, pH value was 6.5, time of hydrolysis was 4h, input amout of enzyme was 1250U/g. Optimization hydrolysis craft condition of protamex was as following: temperature was 50℃, pH value was 7.0, time of hydrolysis was 4.5h, input amout of enzyme was 850U/g.2. Through comparing the effect of the peptone by different enzynolysis Tilapia surimi, results showed that: the peptone yield was more higher when hydrolyzed by AS.1398 neutral protease and papain, were 12.63 percent and 13.25 percent. And the color of freeze-dry product was better.3. Results of central composite orthogonal design of dual enzymatic hydrolysis tests showed: the optimized equation is: Y=13.50616-0.206754X1+0.80289X2+1.294746X3- 0.651231X12-0.8125X1X2-0.0275X1X3-0.126204X22-0.3525X2X3-0.310052X32. This model was able to predict the yield of peptone in the the scope of the experiment. The method of contribution rate showed that: the role of factors on the yield of peptone followed by: input amout of enzyme > time of hydrolysis > temperature. Ridge analysis showed that optimization hydrolysis craft condition of dual enzymatic hydrolysis was as follow: temperature was 56.5℃,time of hydrolysis4.7h, input amout of enzyme was 1174.0U/g. 4. Amino acid composition analysis of dual enzyme hydrolysate was hold. Result showed that: there were full range of essential amino acids and high content of essential amino acids in the hydrolyzate.And essential amino acids accounted for 55.09% of total amino acids. Free amino acids of Tilapia dual enzyme hydrolysate had a high nutritional value.
