Abstract/Summary:
Cephalosporin C(CPC),β-lactam antibiotic, is one of the most important antibiotics in the world and its ratio in antibiotics is arising year by year. In this study, we selected the Mutation Strain with High Productivity by natural screening,UV-mutation and resistance screening from Cephalosporium acremonium HB-2 as original strain, We studied the fermentation process of Cephalosporin C were studied, the result as follows:1,High-yield strain was screened by UV-mutation, which Cephalosporin C synthesis capacity was increased 18% in compare with untreated strain.2,The seed and fermentation medium were optimized through orthogonal design experiment, and the effect of seed age, inoculation volume, DO, fermentation temperature, feed-batch progress on Cephalosporin C synthesis were also studied.The synthesis capacity of Cephalosporin C reached 25g/L on the optimization conditions of fermentation process.3,The change of broth mycelium shape was observed in the fermentation process. The effects of the instant carbon sources and nitrogen source, methionine and inorganic salt on CPC biosynthesis were studied. Biosynthesis of CPC was repressed when instant carbon and nitrogen sources were up to high concentration. Higher concentration phosphate salt inhibited from the biosynthesis of CPC. Methionine can promote metabolism and mycelial differentiation, which strengthen biosynthesis of CPC. When the mycelial has got to a certain extent differentiation, methionine concentration has nothing to do with the biosynthesis of CPC. Namely, the mechanism of methionine is independence with its concentration.4,Example for 60m3 fermentation pot,the HALF-continuum fermentation process of Cephalosporin C were ameliorated and optimized,and were succeed,the fermentation index of Cephalosporin C was increased 20% in compare with the old。...
