This dissertation mainly dealt with the extraction and separation conditions of active components of litchi fruit seed, and sorts and contents of amino acid and polyphenol in it. The results obtained laid theoretical foundation for large-scale extraction, purification and realization industrialization of active components from litchi fruit seed.There are 12 kinds of free amino acid in Litchi fruit seed extract diluted 100 times were detected by ion chromatography, among of these, the relative content of threonine, arginine and glycin was 64%. And there are three of the 12 were unknown and volatile , the anticipative separation effect of ion exchange resin did not come true. Ion chromatography could detect the free amino acid in Litchi fruit seed and the process of separation and purification effectively.The content of free amino acid of litchi fruit seed was detected by TNBS: the content of free amino acid of litchi fruit seed of water extraction is 1.526mg/g, and which of ethanol extraction is 1.860mg/g. The results showed that ethanol extraction is better than water extraction. The synthesis of p-methyl benzene sulfonic chloride and valine derivative obtained laid theoretical foundation.Different concentration separated products were isolated from ethanol extract of litchi fruit seed by AB-8 macroporous resin, then further separation and purification steps that were done through chromato graphyon SephadexLH-20 column were used to 10% and 30% segment, and the separated products were determined by IS and NMR. The results showed that the high activity segment include polyphenol.The sorts and contents of polyphenol which in different separation parts and different kinds of litchi fruit seed were determined by HPLC. It showed that the contents of chlorogenic acid in 10% segment was 689.77mg/g, the contents of chlorogenic acid in 30% segment were 403.91mg/g, the contents of p-coumaric acid and rutin in 45% segment were 3.132mg/g, 56.173 mg/g.There are plenty of polyphenol in ethyl acetate extract, which from kernels of 5 kinds of litchi and Chinese medicine litchi chinensis, the contents of caffeic acid and vanillic acid were 0.5667mg/g in Nuomici, protocatechuic acid was 0.5085mg/g in Guiwei, catechin was 0.1034mg/g in Baila, caffeic acid and vanillic acid were 0.1730mg/g in Rouhebao, catechin was 0.1034mg/g in Chinese medicine litchi chinensis.
