| In this scientific dissertation,soil,mine water,coal and biochemistry water sample were collected from Guizhou,Sichuan,Shanxi,Shandong,Liaoning,Jiangxi provinces respectively,which could obtain the desulfurize bacteria.About 28 strains were isolated, and six strains that from different origin were chosen to be identified through morphological and molecular biology technique,and biological characteristics of them were studied.The results were shown as follows.1.By using 9K culture medium,about 28 strains were isolated,six active strains that from different origin were chosen to be studied.They were SCJ3,LDK1,JLS1,SDJ2,SXT3,SYT4.2.Identified by physiological technique,the strains SCJ3,LDK1,JLS1,SDJ2,SXT3,SYT4 all had the following results:The thalli shape of the strains were short bacilliform,capsule could be seen,gram staining was negative.They could grow autotrophically with Fe2+,S0 and glucose respectively as sole energy source.However, it could not grow with peptone.Among them,the strains JLS1,SDJ2,SXT3 could grow with Na2SO3.However,the using efficiency of Fe2+ and S0 was stronger than Na2SO3.The optimum growth temperature of isolated six strains was 28~30℃,and the optimum growth pH value was 2.0~2.5.3.Identified by physiological,sole energy source using efficiency and molecule biology technique,the strains JLS1,SDJ2,SXT3 were Thiobacillus.ferrooxidans,the strains SCJ3,LDK1,SYT4 were Acidithiobacillus.ferrooxidans.4.The oxidation of Fe2+ and S0 was quite strong of SCJ3,LDK1,JLS1,SDJ2,SXT3. |
