CoQ10,which is also called ubiquinone,is an important transferring-hydrogen carrier of biologic respiratory chain,so it has a significant influence on human beings.At present,it has been widely used in the treatment of cardio-cerebrovascular disease,hypertension,nervous system and skin diseases.In the field of cosmetic products and health products etc,it is also used.The test of production of CoQ10 by microorganism fermentation was carried out.It was described from two aspects of extracting CoQ 10 from PSB, study on analysis methods and selection of PSB,the following fermentation.Ultrasonic extraction and two other assistant methods—zymolysis and freezing & thawing were used for cell disruption.According to the test, Ultrasonic extraction with the help of freezing & thawing was considered to be the best method because of its high CoQ10 extraction amount.It was proved that visible spectrophotometry,UV spectrophotometry and HPLC were all useful methods for the determination of content of CoQ10,but visible spectrophotometry was the best one.It helped us build a quick and easy way in the selection of high yield strains. A high yield strain of 1.28mg/gDCW was selected through enrichment,purification and CoQ10 content determination of PSB.Focus on this strain,a series of experiments were carried out to make out the optimum conditions of strain growth,such as the growth temperature,light intensity, start pH,salinity and so on.Finally,mutagenic treatment was done to find out the positive mutants,and a strain with genetic stability showing 60.1%of increasing of the yield of CoQ 10 was obtained.
