In this study,50 male and female mices 100 sanitary KM mices of 3 weeks were randomLy divided into 5 groups.Each group exposure to different arsenic drinking water,the arsenic concertration is 0,0.05ppm,0.1ppm,0.2ppm,0.4ppm, respectively.They were supplied enough drinking water everyday.After exposed to arsenic for 20 weeks,all of the unpregnancy female,pregnancy for 9 days female and male mices were killed and then uteruses,ovaries,embryoes testicleses were collected.Total RNA were extracted from these tissues,then the pure and content were tested.Total RNA were reverse transcripted to cDNA, the cDNA acted as template in Real Time PCR(RT-PCR).ER and PR expressions level of different tissues were analysed by RT-PCR.The possible mechanism of reproductive toxicity exposed to arsenic was explored due to the study results.The experimental results show:(1)The ER and PR expression in uterus and ovary of unpregnant female mices were tested. ER and PR expression is the highest in control group but ovary PR expression is the highest in 0.05 ppm group,and the difference is obvious among different groups(P<0.05). These results show that As2O3 has some influence on ovary ovulation and blastocyst nidation in unpregnant mices.(2)For pregnancy female mices,in ovary, the highest ER and PR expression in ovary is control and 0.1 ppm group repectively,the difference is obvious among different groups(P<0.05). The highest ER and PR expression in uterus is in 0.2 ppm group but there are no difference among different groups(P>0.05).The highest ER and PR expression in embryo is in 0.1 ppm group,the difference of ER expression is obvious between control and the other exposed group(P<0.05),but there are no difference of PR expression among different groups(P>0.05).There results indicate that As2O3 increase the risk of miscarriage in pregnant mice and has feminization effects on male-embryo.(3)For male mices,the highest ER and PR expression in testicle is 0.2 ppm group and 0.1 ppm group,ER and PR expression in control group is lower than the other exposed group,but there are no difference among different groups(P>0.05).The results show that there is no reproductive toxicity on male mices under the experimental exposed dosage.The results of these experiment reveal that As2O3 has estrogen-like effects and reproductive toxicity,and the reproductive toxicity effects correlate with exposed to As2O3 dose..
