The dried root of Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook. f.var.formosana(Boiss.)Shan et Yuan,as a usual chinese medicine is registered in Pharmacopoeia of People's Republic of China(2005 edition)volum 1.An optimum method was established in this paper to extract and purify the effective fraction in Angelica dahurica.HPLC and UV were applied to evaluate the quality of the effective fraction.1.Study on alcohol extraction process of Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et Yuan.Adopting the method of orthogonal test and using the orthogonal table of L9(34),take the content of total coumarins and the ratio of dry extraction as target,investigating the effect of alcohol concentration,alcohol amount,extractive time on the extraction results,and then the optimum conditions for the extraction of Angelica Dahurica was determined,which was adding eight times of 75% alcohol,refluxing and extracting for 3 h,totally extracting 3 times.2.Study on the purification process of Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et Yuan.Taking the content of total coumarins and retention rate as target,acid precipitation technology,acid precipitation technology and extrating with one and two organic solvent were operated in this paper to purify the extract.After investigating the main factors that effect the purification process,the optimum method was established which was extraction with two times petroleum benzene one time,and then raffinate was extracted with 2 times ethyl acetate three times.3.Study on quality standard of the effective fraction from Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et YuanFirstly,HPLC and UV were applied to evaluate the quality of medicinal materials of Angelica Dahurica and the effective fraction in Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et Yuan. The method was established in this paper.And the results show that the average content of total coumarin and imperatorin in 10 batches of effective fraction were 55.849%and 85.779%.Two methods were simple,accurate and can provide a quantitative basis for evaluating the quality of the effective fraction completely. Secondly,heavy metals,arsenic salts,organic residues extracts in the effective fraction were checked in this paper,and the results show that the effective fraction we prepared was safty and reliable.Finally,To further study the relationship between clinical effect on analgesia and chemical constituents.GC-MS was applied to analysis and identify the constituents in total coumarins,and peak area normalization method was used to calculat the content of each constituent.There were 11 constituents were detected by GC-MS.The main constituents were oxypeucedanin imperatorin and isoimperatorin and which content were 10.23%,6.58%and 5.16%.The method was good,and the resolution of sample was reasonable.4.Study on HPLC fingerprint of the effective fraction from Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.var.formosana(Boiss.)Shan et YuanThe best chromatographic condition of HPLC fingerprint was established through investigating the main factors that effect the resolution.Then 10 batches of effective fraction were analysised by HPLC fingerprint,and 14 common peaks on the HPLC fingerprints of the effective fraction in Angelica Dahurica were indicated,taking imperatorin as the reference peak.And then calculating the similarity.degree.The result shows that the HPLC fingerprint of different groups of the effective fraction in Angelica Dahurica have good similarity degree.
