Preparation Technology And Quality Standard Of Gan'anling

Phyllanthus urinaria L.is one of the traditional Chinese medicines and the wholeplant has the function of clearing heat and protecting the liver, removing dampnessand promoting detoxication in folk. The research on tannins indicated that they arethe effective anti-HBV components in Phyllanthus urinaria L.in modernpharmacology. In the work, with the content of tannins as the researching target, thePhyllanthus urinaria L. extract was prepared by a series of extraction and purificationand made into Gan'anling dispersible tablets.On the premise of the best extraction solvent, the optimum extraction process wasinvestigated by L₉(3⁴) orthogonal design as the content of tannins, and the optimumtechnical condition was adding 12 times of 50% acetone, refluxing and extracting for4 times, one hour each time.To select the optimum purification process, the macroporous resin HPD100 waschosen with the optimum absorption condition as following:the weight ratio ofsolution and raw material was 3:1, the ratio of diameter and height of thechromatography column was 1:7, the weight ratio of raw material and resins was 1:1.A bed volume of distilled water and one bed volume of 15% ethanol were used towash away the impurities, and the 70% ethanol was collected, concentrated and driedto be the target extract.The preparation technology of Gan'anling dispersible tablet, Phyllanthus urinariaL. extract 80g, MCC 60g, CMS-Na 28g and PVPP 30.6g, micropowder silica gel 1.4g,and 30% ethanol is added as the moistening agent.In the quality control study, TLC was used for the identification of Phyllanthusurinaria L. The contents of total tannins were determined by UV. The contents ofcorilagin were determined by HPLC. The method of determining the dissolution rateof corilagin in the tablet was also discussed. All the methods can be used for thequality control of Gan'anling dispersible tablets.In view of the low biological availability of traditional preparation, dispersibletable is prepared with Phyllanthus urinaria L. extract which has a clearpharmacologya and all the work provide corresponding foundation for exploitationand research of pharmacodynamic action of Phyllanthus urinaria L.
