| The great quantities of using pesticide cause the serious pollution by hexachlorobenzene(HCB). Now, there aren't economical and effective techniques of the treatment of HCB which have widely applicable area. Otherwise, the biologic treatment, degrading and translating HCB by microorganism has enormous potential. There isn't any report about the biodegrading of HCB in china.Research objective:selected the anaerobic microflora dechlorinating HCB effectively by selective culture medium from polluted environment, and studied on the characteristic of biodegradation of HCB at different condition and the variety of some important parameter during the degradation.Research content and result:(1) Isolation of anaerobic microflora dechlorinating HCB. The sample was the sludge in barrel drain of a chemical plant in Wuhan, which had been domesticated in nature. Selected the anaerobic microflora dechlorinating HCB effectively from the sample using selective culture medium.(2) Study on the characteristic of biodegradation of HCB. Study on the ability of degrading at different infective factor(T,pH,CHCB) . The results were that: The microflora showed notable differences of degrading ability at different temperature, grew well from 20℃to 35℃, with the optimal one of 30℃. The adaptable range of initial pH was from 6 to 9 with the optimal one of 9. HCB was nearly degraded entirely at the concentration of 1mg·L-1, the rate was 45.4% at 10mg·L-1, and 60mg·L-1 was endurable.(3) The variety of some important parameter during the degradation. measure the parameter CHCB,pH and OD during the degradation. The results were that: The anaerobic microflora increased bloomy during the first 14 days of inoculating. HCB was degraded efficiently within the first 7 days. Otherwise, the result told us a discovery, the trend between curves about pH and concentration of HCB is consistent.(4) The biologic diversity of the microflora. Application of 16S rDNA- restriction fragment length polymorphism screening approach analyzing enrichment revealed 5 clusters, and 3 major clusters were sequenced.Reaserch conclusion: the research selected the anaerobic microflora dechlorinating HCB effectively. It belongs to liking warmth microflora of liking warmth bacterium, and the adaptable range of initial pH for it was widely. Then, the microflora had higher ability of activity and endurance. the curves about pH almost could show the dechlorination instead of the concentration of Cl-. HCB was degraded efficiently within the first 7 days. And, there were three major bacteria in microflora at least. |
