The ginkgolides are selective platelet activating factor antagonists and widely applied in the illness of blood vessel of heart and brain. However,in the current market ginkgolides medicine preparations are mostly traditional drug delivery methods and systemic distribution. The partial blood medicine density is low. They can not achieve the verygood treatment result. So it is very important to target more specifically the ginkgolide in the brain by the development in targeted drug delivery systems(DDS).The HPLC-ELSD internal standard method is applied for the determination of GA. the average extraction rates are 82%~85.7% at three different concentrations and the RSD of inter- day is 2.67%.The study attributes no impurities that endogenous interference, so as the GA-PBCA-NP, therefore this method is suitable for the determination of GA. Under this condition,we inject GA in the mice,and the drug concentration reaches its peak in the brain after 30 minites .At this time,the brain absorption rate is 0.513%.Then in the preparation of GA-PBCA-NP,the interfacial polymerization method is better than emulsification polymerization method. When joined stabilizers, the results showed that the products were spherical with the anverage diameter of 200nm, the anverage ebeding ratio of 73.87 % and the mean drug loading of 22.23% in the system of pH 2~3.At last,the products was injected into the tail vein of mice,and we detected that the TA was 1.855. The experiment result indicates the GA-PBCA-NP is brain targeting initially.
