In recent years,some developed countries have set a number of technical trade barriers for imported aquatic products from China due to some antibiotics like chloramphenicol(CAP)were detected in the fishes.In order to overcome the barriers, it is of great importance to develop sensitive analytical approaches for detection of CAP in fishpond water sample and fish sample.Since CAP is usually presents at a very low concentration and various components coexisting in these samples, preconcentration and separation procedures prior to the instrumental determination is a key step in the analysis.In this thesis,two new methods for analysis of trace CAP in water and fish were developed,which are sensitive,accurate,simple,rapid and less reagent consumption. The thesis contains the following four parts:1.The CAP's structure and characteristics are summarized,and some traditional means for analysis of CAP are introduced in the thesis.Besides,the theoretical bases, operation modes,influencing factors and applications of microwave assisted extraction(MAE)and liquid phase microextraction(LPME)are also presented.2.The method for analysis of CAP in water using gas chromatography(GC)with electron capture detection(ECD)was investigated.Was selected as derivating agent, and the optimized parameters were:derivating temperature of 60℃and reaction time of 30min.The detection limit of CAP was found to be 17.62μg/L.3.A new technique for analysis of CAP in water was proposed,which was based on the combination of hollow fiber liquid phase microextraction(HF-LPME)and gas chromatography.C8H18was used as extractant for CAP derivative from water.Various parameters were investigated in detail,including temperature,stirring time,stirring rate and salt effect.The recovery was found to be 99.56%with RSD(n=10)of 2.80%~7.91%,and detection limit was 1.55μg/L.The CAP content in water samples from two fish ponds in Panyu district,Guangzhou,was analysed by the proposed method.4.A new technique for analysis of CAP in fish was proposed,which was based on the combination of MAE,HF-LPME and GC-ECD.Water was used for MAE of CAP in fish sample,using a domestic microwave furnace.The affecting factors were examined.Some valuable initial results were obtained.The recovery were 93.2%with RSD(n=5)of 4.46%,and detection limit was 0.288μg/kg.
