In this paper,the synthesis method and biologic active of 2-substituted porphyrins and their transition metal complexes were studyed. Further more, 2-Schiff base substituted porphyrin were synthesized by the reaction of 2-substituted porphyrin and aldehyde and its transition metal copper complexes were also synthesized . The main work are as following:Chapter 1st the synthesis method and especial character of 2-substituted porphyrins were summarized, and the development of Photodynamic therapy (PDT) to cancer were summarized too, in which porphyrins were used as photosensitizes.Chapter 2ed 2-nitro-substituted porphyrin (2-NO2-TPP, 3) and its copper complex (Cu(2-NO2-TPP), 1) and nickel complex (Ni(2-NO2-TPP), 2) were synthesized by two different synthesis methods. These complexes were characterized by element alanalysis, infrwered spectra, UV-Vis spectra, mase spectra, 1HNMR.The "one pot boiled" method was adopted to synthesize 2-nitro-substituted metalloporphyrin after compared two different . Further more, 2-nitro-substituted porphyrin was obtained by removing the transition metal ion from metalloporphyrin in the condition of thick vitriolic acid .Chapter 3rd 2-amino-substituted porphyrin (2-NH2-TPP, 4) was synthesized by two different methods and then its transition metal Co,Ni,Cu,Zn complexes (5), (6), (7), (8) were also obtained. They were characterized by element alanalysis,infrwered spectra,UV-Vis spectra,mase spectra,and 1HNMR, etc.Chapter 4th The antibacterial activity of 2-amino-substituted porphyrin (4) and 2-amino-substituted porphyrin cobalt complexes (Co(2-NH2-TPP), 5) were studied. It is found that, 2-amino-substituted porphyrin (4) don't have antibacterial activity,but its cobalt complexes (5) has antibacterial activity. The anti-tumour activity of the 2-nitro -substituted porphyrin copper complex (1) and 2-amino-substituted porphyrin (4) were also estimated. By detecting the survival rate of the tumour cells under the action of those complexes ,we found that, the anti-tumour activity of 2-amino-substituted porphyrin (4) was stronger than that of 2-nitro-substituted porphyrin copper complex (1).The reason was also given.Chapter 5th 2-Schiff base-substituted porphyrin (10) was synthesized by the reaction of 2-amino-substituted porphyrin (4) and 4-((3-phenyl oxyphenyl)-propyloxyphenyl) phenyl formaldehyde and its copper complex (11) was obtained by adding copper salt in the solution of (10). The anti-tumour activity of 2- Schiff base -substituted porphyrin (10) was studied, and the result showed that the anti-tumour activity of 2-Schiff base-substituted porphyrin (10) was stronger than that of 2-amino-substituted porphyrin (4) ,and was stronger than that of 2-nitro-substituted porphyrin copper complex (1) too.
