Due to PNIPAAM with good temperature-sensitive property but few biocompatibility,by using free radical polymerization,PNIPAAM were saccharified. saccharide compoud including Dextran and Galactose were seleced,and then temperature-sensitive and biocompatibility hydrogels scaffold were prepared.By controlling the change of temperature,the target of detaching cell naturally is looked forward to achieve without enzyme,and the function of cell will be protected perfectly.In this paper,PNIPAAM were saccharified by using dextran firstly:glycidyl methacrylate(GMA)derivatized dextran(GMA-Dex)was synthesized by modifying dextran chemically by using GMA,through copolymerization of GMA-Dex with NIPAAM in aqueous solution.A series of novel hydrogels p(NIPAAm-co-GMA-Dex) with different weight ratio(0.2,0.4,0.5,0.6,0.8)were prepared.Both GMA-Dex monomer and p(NIPAAm-co-GMA-Dex)were characterized using FTIR spectra and ~1H-NMR,the influece of r on the LCST and swelling ratio were studied.The study results show that p(NIPAAm-co-GMA-Dex)gels are temperature-sensitive in aqueous and in DMEM.The lower critical solution temperature(LCST)in DMEM lower than in aqueous,the deswelling speed and reswelling speed higher in DMEM than in aqueous.Hydrogels were applied in cell culture.Fibroblast L929 were cultured on p(NIPAAm-co-GMA-Dex)gels.cell morphology were observed by phase-contrast inverted light microscopy and scanning electron microscopy(SEM),L929 exhibited good morphology;cell proliferation curve and MTT indicate that dexran promoted cell gowth.The results show p(NIPAAm-co-GMA-Dex)gels are better than PNIPAAM gels in biocompatibility.cell detachment results show that cell can be detached naturally by reducing temparature while avoiding destroying the function of cell and protecting the cell perfectly.In vitro HepG-2 were cultured P(NIPAAM-co-GAC)hydrogeis,the results indicate that Galactose promote HepG-2 proliferation rapidly,P(NIPAAM-co-GAC)hydrogels are also biocompability.The more Galactose are,the better the cell proliferate.
