In this paper, the extraction, purify and determination of resveratrol from grape seed ultrafine-powder had been studied. All of these provided reference for the foundation of determination standard of resveratrol from grape seed ultrafine-powder.The process of ultrasonic abstraction followed: taking 1.000g grape seed ultrafine-powder, joining 50mL methanol, under conditions with ultrasonic frequency for 40khz,abstraction temperature for 60℃,abstraction time for 10min,abstraction 4 times ,the material was ultrasonic abstracted , merging extracting solution , freezing gentrifugalism , taking supernatant,revolving evaporation to dry. The process of organic-extraction followed: the dry matter was dissolved by 100mL CH3COOCH3:H2O=1:1 solution. collecting ethyl acetate layer,decompressing condense to dry, constant volume to 10mL measuring flask by methanol.After the methodology of HPLC and the ultraviolet spectrophotometer were inspected, the results showed: the accuracy of HPLC instrument was good, the RSD of accuracy was 1.173%, the RSD of repeatability was 2.043%, the recovery was between 93.70% to 98.68%, and the average recovery was 97.13% and the RSD was 2.113%, the average content of Res was 8.482μg/g, the linearity range was between 0.2μg/mL to 10.0μg/mL. But using ultraviolet spectrophotometer, the RSD of accuracy was 2.221%, the RSD of repeatability was 2.644%, the recovery was between 87.90% to 97.60%, the average recovery was 93.37% and the RSD was 4.725%, the average content of Res was 8.009μg/g, the linearity range was between 0.2μg/mL to 1.0μg/mL.The results of significance analysis to two determination methods shows: they have extremely significance difference. All of these results indicate: the determination of Res from grape seed product using HPLC is better.The HPLC conditions were: shim-park VP-ODS(250×4.6 I.D) column with acetonitrile-water- acetic acid (CH3CN: H2O: CH3COOH =28:70:2) as the mobile phase and detached at the speed of 1.0mL/min and the wave length of 305nm and the temperature of column was 30℃and the sample size was 20μl.
