| In this paper, the extraction, isolation, purification, structure and bioactivities ofthe polysaccharide from fruit bodies of Agrocybe aegerita was studied. The optimumconditions of extracting polysaccharide from A. aegerita were determined throughsingle-factor experiments and orthogonal experiment with ultrasound-assistedextraction method.The optimum conditions were: ultrasonic time 30min, ultrasonicpower 320W, extracting temperature 50℃, extracting time 3h. Under these conditions,the extraction rate of crude polysaccharides reached to 19.44%. After degreasing,removal proteins and dialyzing, the polysaccharide was applied to Sephadex G-100and DEAE-Sephadex A-25. Then we got a purified polysaccharide fraction ACPS. Thepurity of ACPS was confirmed by PAGE and HPLC. The chemical structure wasanalyzed by TLC, HPLC, UV, IR, 1HNMR. The results showed that ACPS was anacidic polysaccharide consisting of D-galactose, L-rhamnose and uronic acid residueswith a molecular mass of 11 863, and the configuration of ACPS wasβ-glycosidic.The effects on scavenging·OH and·O2- of A. aegerita polysaccharides were studiedby Fenton system and Pyorgallol Autoxidation Method respectively, which werecompared with the effect of Vc. Under the condition of mimic gastric juice, the activityof A. aegerita crude polysaccharide to scavenge NO-2- was eliminated. The spleen indexand thymus index, the phagocytic rate and phagocytic index on mice macrophageswere detected for investigating the immunological activity of A. aegeritapolysaccharide. Anti-tumor activity of A. aegerita polysaccharide on S180 mice wasalso studied. |
