| 6-(10-Hydroxydecyl)-2,3-dimethoxy-5-methyl-l,4-benzequinone(Idebeno ne) came into the market at Dec. 1986 which is the fist drug to cure Friedreich's Ataxia(FRDA). It is used to improve cerebel disorder in the clinic. Datum on the value of antioxidant show that Idebenone is the most effective antioxidant for external application, it have been to used to anti-age. Idebenone has extensive application foreground at medication and health protection, it has largrly vendibillity. From so on, Idebenone has been producted by traditional technology which is low yield. Improving the technology and increasing yield are main study in this paper.Fist study on the intermediate, 10-acetoxydecanoyl chloride, was synthesized with 10-hydroxyldecanoic acid, acetic anhydride and acylating agent via acetylation and chlorination. Idebenone was synthesized with 3,4,5-trimethoxytoluene and 10-acetoxydecanoyl chlride via Friedel-Crafts reaction, alcoholysis, reducing reaction, oxidation raction and alcoholysis again. The overall yield was about 32%. The structures of the compounds have been confirmed by GCMS.A lot of experiments and some improvements are made in this paper. In study, PCl3 and SOCl2 which are found to effective, easy to purification and high yield for chlorination replace PCl5 as chlorinating agent. In the Friedel-Crafts reaction, studying on the way of charging up, temperature of charging up, reaction reagent, so reducing reaction time largely and making sure high yield. In the alcoholysis, we study on yield with different bases, and improve reaction conditions. Fremy's Salt is used as oxidant at oxidation reaction in traditional technology. Fremys Salt which is difficult to make and save, is replaced by CAN, so it solves the problem about oxidant. After alcoholysis, the products are edulcorated by Flash Chromatograph and recrystallization. Study on quantity of solvent, and confirm the least measuring flask which receives mother liquid. And discuss way of recrystallization.By comparison with the original material, many improvements are made on technology, production cost, operation and so on. |
