| Environmental hormones, also was called environmental endocrine disruption chemicals, acted as estrogens in human and animal bodies. Most of them were discharged into the environment with the production and the life of the humanity. It had been found that environmental hormones existing in the environment having adverse effects on endocrine systems in human and animals and led to reproductive, developmental and behavioral disorder. Of all EDCs, bisphenol A (BPA) was very important one. With BPA used in great quantity, it was detected in all kinds of waters all over the world. The BPA could be used as aerobic microorganisms' carbon resource, and could be degraded into in-poisonous substances, so it was an effective method to use microorganisms in degrading waste water. Some correlative work was done:Fist of all, isolation of BPA degrading strain. With the classical technology of screening, bacteria high-effectively degrading BPA could be obtained, which only take the BPA as carbon and energy sources in the saturated solution of BPA. This kind of bacteria could degrade the BPA effectively. Secondly, bacterial strain Identification. The traditional classification and 16SrDNA gene sequence analysis methods were combined to identify the strain. The result indicated that the physical and chemical standard conformed to the characteristic of Pseudomonas. From the contrast analysis of 16SrDNA gene sequence, the strain was found that it had homology property with Pseudomonas by 99%, the strain could be cognized Pseudomonas.Thirdly, study of the cultivation condition of strain P-04. The carbon sources, nitrogen sources, aeration rate, original pH and temperature, etc., which influenced the growth of strain and the degradability to BPA, were discussed here. The optimum of conditions were pH7, 50mL cultivation medium in 250mL shaking flasks, 30°C.Fourthly, ultraviolet radiation experiment. The BPA degrading rate of cream-colored, stable generated strain P-24, induced 45 seconds from strain P-04, was 87.6%, which was increased by 4% than that of P-04. The strain P-24 had better tolerance and its degrading process had little change than original strain.Fifthly, study of fermentation. Batch cultivation of strain P-04 was carried out for applying the bacteria-degradation of BPA to industry, and three models of the development of bacteria, the consumption of carbon and the production of degradation agent were obtained according to the experimental datum. Finally, the preliminary application of BPA degradation. The active sludge had better removing rate by 19% of COD of Tianshan waste water than that of strain P-24, but it could not degrade BPA. The strain P-24 had higher COD removing rate by 33%, and BPA removing rate by 45% than active sludge in BPA waste water. The strain which had been domesticated and isolated had better degrading effect on BPA in BPA waste water. The mix strain had higher COD removing rate by 31 % and BPA removing rate by 35% than P-24, and 2 times, 20 times than those of active sludge.These research results will provide a foundation datum for treating of environmental hormone BPA and finding degradation gene of the bacterium. |
