The Chinese mitten-handed crab always lives in three kinds of environments: rivers, lakes and ponds. Heavy metals evidencely influence its growth. They accumulate in the crabs and do harm to people's health through food chain. Along with the escalation of crab breeding, more and more attentions have been attracted. This article researched the accumulation and distribution rules of heavy metals and the inspection methods in the mitten-handed crabs.With the technology of microwave digestion and hydride generation—atomic fluorescence spectrometry, a content inspection method of As and Hg in crabs were established. The operating conditions of the instrument were optimized. The effects of reducing agent and reducing shelter on the detection of As and Hg were investigated. In addition, the accuracy and precision were also validated, which was of great instructional significance in routine analytical works. This method was sensitive and suitable for content inspection of As and Hg content in crabs.Moreover, an inspection method of Pb and Cd was studied. The detection limit of Pb was 1.30μg/L, and Cd was 0.052μg/L. The calibration curve of Pb was linear in the range of 0.0~50.0μg/L and the equation was: Y=0.0012x+0.0038, (?)=0.9932. The calibration curve of Cd was linear from 0.0 μg/L to 10.0 μg/L and the equation was: Y=0.05951x+0.026, (?) =0.9945. The intra-assay variation coefficients of Pb was 2.86%~8.89% and the inter-assay was 2.70%~10.00%, while the intra-assay variation coefficients of Cd was 1.35%~8.00% and inter-assay was 5.48%~9.09%. When the crab samples are spiked with 0.15μg/g, 0.30μg/g and 0.45μg/g of Pb and Cd standard samples, the average recoveries of Pb were 82.04%, 94.94% and 110.22%, and the Cd were.47%, 0.32% and 88.91%. The statistics adequately showed that it was a sensitive and suitable method for the determination of Pb and Cd contents in crabs.What's more, the accumulation of heavy metals in crabs of three breeding patterns (rivers, lakes, and ponds) were researched through the detection of As, Hg, Pb, Cd, Cr, Cu, Zn in the tissues of branchia, muscle, liver (sex gland), and the water and sediments...
