Polygonum cuspidatum siob et Zucc.is one of Chinese traditional medicines in common use with its radix and rhizome,which is a member of Polygonum Linn.This species is abundant in natural resources,with wide distribution in Sichuan, Hubei, Shanxi et. Resveratrol and polydatin,as important constituents of P. cuspidatum are found to pose many specific pharmaceutical activities such as antineoplastic,anti-oxidation,natable cancer chemopreventive activity,reducing the risk of coronary heart disease(CHD),et al. According to known papers,the resveratrol content in P. cuspidatum was the highest among all studied plant species,so its extraction becomes a research focus.In this work,three different extraction techniques were used for the same herb sample to evaluate their extraction efficiency,the extract was determined using high performance liquid chromatography(HPLC).Additionally,the contents of resveratrol and polydatin of 5 samples from Sichuan were determined by the optimal method above.Several main results obtained are summarized as follows:1. The efficient of three extraction techniques,alcohol extraction method,water extraction method and supercritical CO2 extraction were studied.It was found that alcohol was the best extracting solvent.Meanwhile,according to the results,the best experiment condition was 95% alcohol as solvent,in 60℃ for 3h.2. The contents of resveratrol of P. cuspidatum samples from 5 regions in Sichuan were determined.The highest and lowest contents of resveratrol were 3.85mg.g-1 and 0.25mg.g-1,with the average of 2.13mg.g-1.For polydatin,the highest and lowest content were 22.9mg.g-1 and 7.69mg.g-1, respectively,with the average of 12.52mg.g-1. |
