Oat is one of main crops in the northwest area. Recently important physiological functions on oatβ-glucan have been found out. Oat oil enriches unsaturated fatty acid. Extraction and purification ofβ-glucan and exploiting functional oil can promote oat value, which will contribute to development of healthy food and medical industry on the basis of oat and accelerate economic development of northwest area to the extent.β-glucan and oil are rich in oat bran. The thesis studied such main factors which affected ratio of extractingβ-glucan from oat bran without oil as extraction time,pH value,temperature and ratio of solvent to solid. Next the optimal technology parameters of extractingβ-glucan were determined by orthogonal table, and then such key processes as removing starch and protein and enrichingβ-glucan to full extent were investigated. Oil was extracted from oat bran by supercritical carbon dioxide. Through investigation the optimal technology parameters of extracting oil were determined and compositions of fatty acid in oil were analyzed by GC-MS.The optimal technology parameters of extractingβ-glucan were temperature 75℃,pH value10, time 2 hours and ratio of solvent to solid 1:17. Starch was decomposed by moderate-temperatureα-amylase and degree of colour change evaluated if starch was completely decomposed after usingα-amylase twice. 20Uα-amylase per 170ml solution was added and reaction lasted twenty minutes before extraction, and then 50Uα-amylase per 100ml solution was added and reaction did twenty-five minutes after extraction. In the case starch was completely decomposed.Ratio of removing protein didn't change any longer after using the way of Sevag six times if experiment was added and here ratio of its elimination was 80.3%.Ratio of removing protein at pH4.5, using trypsinase,using trypsinase before precipitating protein at pH4.5, and by the way of Sevag together with trypsinase was 95.5%,59.4%,95.8% and 71.8% respectively, while ratio of keeging polysaccharide was 83%, 60%, 90.6% and 90.4% respectively. To summarize, after using trypsinase result of removing protein at pH4.5 was the best among them.β-glucan could be precipitated under the condition of different amount of ethanol and 70% ethanol can precipitateβ-glucan to the full extent.
