The paper relates to the study of the synthesis of α-2 adrenergic receptors sedative Detomidine and a novel penicillanic acid suifones P-Lactamase inhibitor Tazobactam.Detomidine is a unique seadative and pain reliever ,introduced as a sedative analgesic for use in horse and cattle in Europeancountries in 1983.detomidinewas developed to provide similar clinical effects as xylazine but without some of the associated side effects.We research it's two synthesis route in this part.In the first route :we start from cheap glycin via a series of reaction: acylation, esteration ,condensation with ethyl formate under base condition.ring-closing with KSCN in presence of HCl.Followed by oxidation with H₂O₂,reduction with NaBH₄ oxidation with MnO₂ yield the important intermediate imidazole -4-carboxaldehyde.then. it was reactived with 2,3-dimethyl-phenylmagnesium bromide,followed reduction with Pt/C.In the second route:we start from facile imidazole:first we synthesis 2,4,5-triiodoimidazole with iodine ,then selective deiodination , followed by via a series of reaction: protection of amido in imidazole,reduction ,nucleophilic addition ,deprotection yield the target product.Tazobactam is a novel p-Lactamase inhibitor belonging to a clas of penicillanic acid suifones. and researched and developed by Japan Dapeng Pharma CO.
