| The polyphenol compound, included phenolic acids and flavonoids, is a kind of biological activity compounds. In this thesis, phenol acids and flavones in Chinese traditional medicine were determined by high perform liquid chromatography-amperometric detector (HPLC-AD). Additionally, photo-diode array detector, pre-column derivatization, chemometrics were also combined with HPLC-AD to determin polyphenol compounds. The studies have important significance in broadening the application range of HPLC and in quality control of Chinese traditional medicine. This thesis consists of six chapters. The works of author as follows:1. To establish an HPLC-AD method for simultaneous determination of the contents of gallic acid, protocatechuic acid, chlorogenic acid, caffic acid, vanillinc acid and ferulic acid in Yiganshukang capsules. The average recoveries of the six phenolic acids were between 95.8 99.8% with RSDs of 2.1 3.2%. The method is simple, rapid, reproducible and accurate. It can be used for the routine analysis of the six phenolic acids in Yiganshukang capsules.2. To establish an HPLC-AD method for simultaneous determination of the contents of protocatechuic acid, protocatechuic aldehyde, caffic acid, quercetin and kaempferol in Ilex purpurea. The average recoveries of these compounds were between 98.6 99.4% with RSDs of 2.3 2.8%. The method is simple, rapid, reproducible and accurate. It can be used for the routine analysis of the herb of Ilex purpurea.3. To establish an HPLC-AD method for simultaneous determination of the contents of rutin and quercetion in Flos Sophorae, Semen cuscutae and Flos Buddlejae. The average recoveries of the rutin and quercetin were between 97.2 99.3% with RSDs of 3.2 4.2%. The method was applied to determination of the hydrolysis rate constants of rutin, which is 8.53 K·min-1 at 368K, 7.20 K·min-1 at 358K.4. To develop an HPLC-DAD-AD method for the determination the contents of chlorogenic acid, caffeic acid, baicalin and luteolin in Sanjing Shuanghuanglian Oral Liquid. The contents of chlorgenic acid, caffeic acid luteolin were determineded with HPLC-AD, and the content of baicalin was determined with HPLC-DAD. The average recoveries were between 96.6 99.6% with RSDs of 2.5 4.1%. The method is simple, rapid, reproducible and accurate, and it can be used for the routine analysis of the four compounds in Shuanghuanglian Oral Liquid.5. Through adding pyrogallic acid to mobile phase, a indirect HPLC-AD method of determination of the non-electroactive Chrysin in Orxylum indicum (L.) Vent was presented. The method broades the application range of HPLC-AD.6. The HPLC-AD combining with a new method of Fourier self-deconvolution (FSD) was applied to the simultaneous separation and determination of chlorogenic acid, caffeic acid, ferulic acid, reveratrol and quercetin and luteolin in Reyanning capsules successfully. Compared with HPLC-AD, the new method is rapid, reproducible and accurate. |
