The in situ gel is a new drug delivery system, which is injected to reach solidification in the target location because of the role of body fluids. Periodontitis is inflammation in gum. In situ gelling is a technology which is suitable for the therapy of Periodontitis. Minocycline Hydrochloride is chosed as the model medicine in biopharmacy, physical pharmaceutics, and other theoretical guidance for the study of treatment of periodontitis by local release preparation.Polymer Carbopol, sodium polyacrylate and cellulose derivative EC are adopted as the main drug sustained-releasing materials.The parpared matrix can release MINO for 5-7 days.The method of preparation analysis is optimized.Dissolution rate is an important factor of influencing internal quality. And the various factors are studied, which influence the gel of sustained-release. Orthogonal project is employed to screen the prescriptions which based on the study of MINO-factor experiment. Verification experiments about the released rate of optimized prescription are done .The results showed that the released rate of sustained-release gel at lh,3h,7h was 1525%,3050% and 60-80% respectively. The f2 similar factor analysis explains that the recur capability of sustained-release MINO gel releasing is good and the craft is stable.Through the study of mass accessorial drug's stability,the result shows that temperature ,pH and the ion of acrylic acid have obvious effects on isomeric change of chirality of MINO.The result of accelerated test indicates that MINO in NOVERITE 7S is unstable,and it is relative stable in CP, EC .By the rounded analysis of the two experiments above, we use EC and CP as the controlled-release materials to optimize the formulation.It has few reports about development of sustained-release MINO gel in china. So it is a original study.The results studied show that it is a good foundation of new delivery system which is used in oral location.
