In this article ethanol-degradation bacteria is deeply studied based on the alcohol-poison-medicine and treatment of waste ethanol pollution at home and abroad. A strain of bacteria which can degradate ethanol well is selected from Jiaoni and Huangshui in the experiment and its biochemical characteristics are researched. The test has supplied reliable technic parameter for the exploitation of biotreatment technique of alcohol poison and waste ethanol pollution.The follow aspects are studied in the paper.1) The selection of ethanol-degradation bacteria.The bacteria sample gotten from Jiaoni Huangshui and Cupei is cultured on the medium containing ethanol. The ethanol-degradation bacteria survived in the first selection is trained in a serious media with different ethanol consistence.A strain of ethanol-degradation bacteria which can degradate ethanol 1.45ml per day averagely is reselected.2) The determination of the optimal cultureing conditions for the ethanol-degradation bacteria. An orthogonal test of three factors and four levers is designed to determine the optimal medium and a single factor test to determine the optimal culturing condition. As the result the optimal medium for the ethanol-degradation bacteria is composed of soluble march 3g, NH4Cl 0.15g, K2HPO4·3H2O 0.1g, NaCl 0.04g, MgSO4·7H2O 0.05g, FeSO4·7H2O 0.001g and phosphate buffer 1ml, microelement 0.5ml,pure water 100ml. Then the optimal growing condition is to inoculate the ethanol-degradation bacteria 107/ml after solubilization oxgene for one day in the medium(pH 7.0) and culture them at 37℃with 160rpm shaking. Additionally the activity of alcohol alcohol dehydrogenase(ADH) is studied in the experiment which can reach 145U.3) Research on growth characteristic of the ethanol-degradation bacteria. Growth curve is described by observing the change of culture solution consistence of the ethanol-degradation bacteria in a cycle.It shows in the curve that the lag phase, exponential phase, stationary phase is 14h, 3h, 9h respectively and after culturing 26h the ethanol-degradation bacteria enter into death phase.It showed in the research that the ethanol-degradation bacteria selected can degradate ethanol 1.45ml per day averagely.
