The tetracyclines generated by actinomyces are broad-spectrum antibiotics that are proved to be highly effective against nearly all Gram-positive and Gram-negative bacteria in clinic application. They can also be used as animal drug,disinfectant, antistaling agent. According to recent reporting they can also be used to cure and diagnose cancer. So, the study on tetracyclines has now become the focus of public attention once more. Fluorescence spectrophotometry has been applied widely in pharmaceutical analysis because of its high sensitivity and good selectivity. In this paper, we have done the following work in the exploitation of tetracyclines' new determination method and interaction druggery and biomolecule.First, new micelle stabilization, sensitized fluorescence methods for the determination of Doxycycline(DC) and Metacycline(MTC) are proposed. We studied the fluorescence spectra of the system and optimized the experiment condition. The results show that the addition of proper surfactant has made thesystem solubilization, stabilization and enhanced sensitivity. The results also show that antimagnetic metal ions sensitize or has no influence on the fluorescence, but paramagnetic metal ions quench the fluorescence. Following the optimally version designed according to the method, we determined medical content in pharmaceutical preparation. The linear range is wide, the detection limit is low, the recovery is within the range of 95 -105 %. The results are satisfied.Second, a new pharmaceutical analysis method using Eu3+ ion as fluorescent probe for the determination of Doxycycline is established. The optimum condition of forming intramolecule energy transfer complex by DC and Eu3+ is attained. The composition of the complex is defined, too. The proposed method has been successfully applied to analysis the drugs of complicated serum and their pharmaceutical formulations. Moreover, we discuss luminescence mechanics.Third, we has successfully settled the problem of spectra overlap of OTC and MTC by making use of new fluorescent technique—"Zero-crossing" Derivative -Synchronous Fruorimetry. Independent determination of these two drugs are usually difficult. A new method for the determination of MTC under the existence of OTC. Following the optimally version, we determined MTC content in mixture sample. The linear range is 0.2 ~ 27.5 ug-mL"1, the recovery is within the range of 94-103 %. The results are satisfied.Fourth, we study the interaction of druggery and biomolecule by fluorescence quenching method. Bovine Serum Albumin is main block, drug is object of the study. Applying fluorescence quenching method and Forster energytransfer technique, we have studied the quenching mechanism of protein and drugs. Stern-Volmer and Lineweaver-Burk curves were drawing. The binding site, the distance of donor-acceptor and energy transfer efficiency were obtained. According to the thermodynamic parameters, we has conjectured that the main sorts of binding force is static-electricity force.
