Based on the established analyzing method of total flavonoids and marker substance -rutin, the extracting and accumulating technology of the flavonoids in the twigs and leaves of Taxus Cuspidata have been studied by employing the content and transfer rate of total flavonoids as index in this paper. The optimum techniques of extraction was investigated through the factorial and orthogonal design, and parameters of purification by using the macroporous adsorption resin was studied. The techniques was established as following: extracted with 70% alcohol for lhr, 2 times, absorbed by AB-8 macroporous adsorption resin, desorbed by water and 70% alcohol. The cotent of total flavonoids came up with 52.57% by means of spectrophotometry, the content of rutin was 4.84% by using RP- HPLC, with the yield of 7.01%.In addition, the HPLC fingerprints of extraction were obtained with twelve common peaks. The similarity of 10 batches were more than 0.99, which showed that the extracting and accumulating technology could provide the stability sample for bioactivity research. It may be practically valuble for the quality control of Taxus cupidata. and its extracts.The extracting and purifying process flavonoids from Taxus cupidata were carried out for the first time. The results of reaserch provoid the basic data for further bioactivity studies and its exploitation.
