| Phytic acid is important natural compound in soybean oil-seed .It and its major breakdown products have been found to benefit to and also been linked with reduced risk of cancer .It is reported that phytic acid can accelerate gene (P53) of restrain cancer to expression and pass multi-approach to restrain blood vessel engendered, consequently interdict blood supply and reduce nutrition transportation for tumour, there is famine state in tumour and tumour become small. So it is perfect medication for resist tumour .Therefore, it was very important to separate phytic acid form plants, which will be beneficial to the development of chemical industry, food science and medicine. At present, ion-exchange method is the most active method on separating phytic acid form plants. However there are many literature on ion-exchange, but there are majority summarize or simple depict ion-exchange process ,the detailed report on choose of resins and effect of exchange factors and the method of static state or dynamic state is less. The paper study extraction of phytic acid form rapeseed cake .The purification is identified by IR and NMR and so on .In chapter 1: the research status of phytic acid was summarized and reviewed, including the distribution in plants, the analysis method and application, the antinutritional factor, the extraction of phytic acid. The technology of ion-exchange was introduced contemporary. In the same time, the target and method of our research were reported.In chapter 2: the elementary extraction of phytic acid was studied. The factors including the extraction solvent kinds, the concentration , the extraction fashion, the extraction equilibrium time, the neutralization solvent kinds, the mixed extraction solvent kinds, the extraction times, the extraction temperature were study. The results showed that 0.5mol/L HCl was the better extraction solvent, the extraction equilibrium was accelerated by mix round and ultrasonic. But the extraction rate could not be enhanced up by means of the mix round and ultrasonic. At the same time we can find that the extraction rate was enhanced slightly by mixed extraction solvent HC1 and acetone. The neutralization solvent such as NaOH and HCl are the better effect.In chapter 3: the ion-exchange for phytic acid was studied. The static and dynamic ion-exchange methods were applied. The influence factors such as ion-exchange resins kinds, pH value, temperature, velocity of flow, concentration of sample were investigated. At the same time, the effect of resins ion models and mixed extraction solvents were examined. The results revealed that weak alkaline ion-exchange resin is more effective than strongly basic ion-exchange resin. D315 ion-exchange resin is the best adsorption effect for phytic acid. at the same time the Cl model was the best adsorption effect for phytic acid on D315 resin. The mixed washing solvent for phytic acid was investigated. It showed that the solvent of acetone(80%) and HCl(0.5mol/L) is the best washing effect. |
