The present work focus on extraction and separation of phytosterols from Elaeagnut mollis.Diels oil first, the condition of extraction of extraction about four factors was studied and the optimum extraction condition were then obtained and high quality product were acquired with this method. The phytosterols were then analysised with chemistal display, thin layer chromatography, silica column chromatography, Gas chromatography and Gas Chromatography-Mass Spectrometry etc. Ten compounds were identified, the results were summarized as follows:1. A system for separation of phytosterol by thin-layer chromatography has been improved: The system utilized silica gel plates and n-hexane/diethyl ether/acetic acid(80:20:1 V/V/V) and chloroform/diethyl ether(9:1 V/V) as mobile phases. In the first and second directions, respectively. The plate are sprayed with 0.5 mg/mL FeCl3 and heated at 110℃ for 2 minutes.2. A method for separation of phytosterol by GC has been established. The sample were determined in DB-5 column, and BSTFA was used as derivatives reagent。 The oven temperature was programmed to begin at 70°C,hold for 1 min, increase to 295℃ at 2℃/min, hold for 10 min, detector temperature was set to 270℃.3. The parameters of wet saponification for Samara oil were optimized by orthogonal experiment: the sample was saponified in boiled water for 1 hour in alkali hydrolyze conditions, then crude phytosterol was extracted as the proporation 3:1, dissolved in ethanol to saturate state under heat, then decompressed distillation on circumrotate evaporation instrument and crystallize out the content of phytosterol is 19.5%.4. The composition of phytosterol in Samara Oil is as follows : β-sitosterol, 82.34%; stigmasta-5,24(28)-dien-3 β-ol, 7.74%; ergost-5-en-3 β-ol, 3.05%; compounds below 1% (relative percentage ) include ergosterol, stigmasterol, 5-chlorostigmastan-3-yl acetate , stigmastanol , stigmasta-7,24(28)-dien-3 β -ol and stigmastan-3,5-diene. Among all the compounds, stigmasta-5,24(28)-dien-3 β -ol, ergost-5-en-3 β -ol, 5-chlorostigmastan-3-yl acetate , stigmasta-7,24(28)-dien-3 β -ol are discovered firstly.
