Abstract: Grasshopper has higher economic value, because there are many functional ingredients in it, such as abundant protein, essential amino acid, vitamin, mineral elements, unsaturated fatty acid et. But little studies on grasshopper were carried over the time. This paper attempted to make a systematic research on composition, classification and extraction of grasshopper protein, and also studied on the extraction, purification and properties of superoxide dismutase -a special functional grasshopper protein. These studies would pay a way to the exploration and production of grasshopper.The results showed that the protein content of grasshopper was high, the composition of amino acid was desirable, and had all kinds of essential amino acids. So grasshopper was a high quality protein resource.The protein of Acrida cinerea (Thunberg) was divided into water-dissolved protein (43.4%), acid-dissolved protein (1.8%), alkaline-dissolved protein (1.9%) and mellow-dissolved protein (0.3%). The content of water-dissolved protein was high, so grasshopper protein was easy to digest and absorb.The results showed that the optimum extraction conditions of Acrida cinerea (Thunberg) water dissolved protein were as follows: the ratio of grasshopper/solvent (g/mL) was 1/15, pH of solution was 7.6, the NaCl concentration in the extractive reagent was 1.0% and the extraction time was 4 hours.Effect of several factors (pH of solution, heating temperature, heating time) on superoxide dismutase extraction was explored. The results showed that it was under the conditions (pH of solution 7.66, heating temperature 45℃, heating time 40min) that extraction efficiency of Oxya chinensis(Thunberg) superoxide dismutase was better.After denaturization by heating, the extraction was purified through DE-52, got 4 protein compositions, detected activity, collected high activity compositions, finally the specific activity was 137.5U/mg pro, the recovery of activity was 41.2%.The superoxide dismutase was identified by SDS-PAGE after purification, the...
