Studies On Accumulation Of Minerals And Increasing Of Production Of Polysacchride In Ganoderma Lucidum Liquid Fermentation Process

Abstract/Summary:
Ganoderma lucidum is a famous medicinal and edible fungus with special nutritional and medicinal value. Many studies show that some components of Ganoderma lucidum such as polysaccharide and ganodenic acid have great pharmacodynamics activities. This paper investigated the process conditions of liquid submerged culture of Ganoderma lucidum.In the research, a Ganoderma lucidum strain of No.22 was selected for accumulating polysaccharide and the optimal medium for Ganoderma lucidum was optimized by orthogonal experimental design. The best medium was found to be composed of 4% potato, 4% wheat bran broth, 2% glucose, 0.2% KH2PO4 and 0.1%MgSO4.7H2O. The optimal fermentation conditions were as follows: culture temperature at 28°C, aerating rate at 1:1V/V/min, stirring speed at 250rpm, 70-94 h culture time and natural pH.The maximum Zn-accumulating rate of Ganoderma lucidum mycelia was 23% when the Zinc concentration in medium was 30ppm and the mycelia growth was inhibited at the Zn concentration of 60ppm. It grew slowly when the Selenium concentration was above 60ppm in the medium.Effects of pH value on the fermentation of Ganoderma lucidum was also investigated. The results showed that the optimum pH value for mycelia growth was 5.4 and the appropriate pH value for exopolysaccharides production was 3.4. When the fermentation pH controlled at 3.4, the maximum yield of exopolysacchrides was 2.534g/L, while it was 1.711g/L without pH controlled. In pH controlled fermentation, the yield of exopolysacchrides increased by 48.1% comparing with pH uncontrolled fermentation. When we kept the initial fermentation pH value at 5.4 for 48 hours then controlled the pH value at 3.4, the yield of exopolysaccharides was 1.844g/L.The production technology of orally liquid of Ganoderma lucidum was also studied.the best conditions of polysaccharide drawn are nine times of water and one hour.
