| The fungal elicitors, which are extracted from the fungi's cell walls or from their culture, have been widely used in the field of cell culture of plants nowadays. They are proved to be multifunctional factors to plants and have the ability of regulating the growth of plants, inducing the plants resistance to pathogens, and simulating the synthesis of plants secondary metabolite. This paper centers on the inducement effect of the oligosaccharins isolated from Fusarium moniliforme, a kind of elicitor, to the cotton and Salvia miltiorrhiza hairy roots. In this process, the polysaccharide was basically isolated from Fusarium moniliforme cell walls by pretreatment with NaOH solutions and H3PO4 successively, and some impurities were removed. IR spectrum of this polysaccharide showed that the polysaccharide chain was linked by β-glycoside-bond mostly. Further Fusarium moniliforme oligosaccharins was obtained by hydrolyzation with trifluoroacetic acid and under high temperature. And fluorophore-assisted carbohydrate electrophoresis(FACE)was used to analysis the oligosaccharins components. The result suggested that the oligosaccharins acquired from any method was composed of four oligosaccharides.Fusarium moniliforme elicitor, refined by hydrolyzation under high temperature, was used to pretreat the cotton seeds, then the effects of the elicitor on the germination of the seeds , growth of the seedling and the activities changes of some enzymes which were relative to resistance were studied. The results showed that Fusarium moniliforme elicitor can evidently inhibit the seeds germination and seedling growth, and induce the β-1,3-glucanase, peroxidase and phenylalanine ammonialyase activities. Furthermore, Salvia miltiorrhiza hairy roots were induced by Fusarium moniliforme oligosaccharins with different concentrations. Then the inducement effect of the oligosaccharins with different concentrations on the hairy roots growth, peroxidase activities and tanshinon content were studied. It was showed that the Fusarium monoliforme oligosaccharins can accelerate hairy roots grow, induce the tip of roots bulge. In addition, the peroxidase activity, tanshinone biosynthesis and the exudation were enhanced via the treatment. |
