This paper studies the synthesis of intermediate of 9-(2-hydroxyethoxymethl) guanine and attempts to find out the best route for the synthesis of 1,3-dioxolane by means of changing the catalyst category and adjusting its usage, reaction temperature, reaction time and the ratio of the raw materials, using trioxymethylene and glycol, thus improving the yield ratio of product to 75%. On the basis of research and experiment, and with the material of 1,3-dioxolane and acetic anhydride, the paper tries to work out the optimal way for the synthesis of 2-oxal-l,4-butanediol diacetate by changing the catalyst category and by the initial reaction condition achieved through orthogonal experiment,?the condition which serves as the basis for further optimizing the reaction condition, thus achieving the yield ratio of 95%. Product(l) may be synthesized by 2-oxal-l,4-butanediol diacetate and N (2) -acetyl guanine. The product yield ratio reaches up to around 60%.
