objective: 1) To study the application of four methods which were JH clarifying agen, chitosans, ZTC1±1 clarifying agent and Ultracentrifuga-tion in the producing technology of Xuanfeiqingre Granules. And those best technology were compared with traditional clarifying method which was alcohol-precipitation. 2 ) To study optimum inclusion process for oil from Pericarpium Citri Reticulatae and others.Methods: 1) Based on the mono factor design, those technology were optimized by using uniform design which took the differences of the extraction rate, the contents of ephedrine and hesperidin treated by those methods and not treated as indices. Those four clarifying methods were compared with alcohol-precipitation. 2) According to the mono factor design, we used the ultizision ratio of oil and the productivity rate as indices to select the process condition.Results: 1 ) Using JH clarifying agent, The optimized technology contained 1:1 water extracts, 2.2% JH clarifying agent, pH 6.4 and 20℃. 2) Using chitosans, The optimized technology contained 1:1 water extracts, 2.2% chitosans, pH7 and 35℃. 3) Using ZTC1±1 clarifying agen(2B: A) The optimized technology contained 1:1 water extracts, 2.4% ZTC1±1 clarifying agen, pH 4.8 and 35℃. 4) Using Ultracentrifu-gation, The optimized technology contained 1:1 water extracts, the rate of stirring is 17000r/min. 5) Alcohol-precipitation is not only greatly reduce the extraction rate (about 21%), but also can maintain the contents of ephedrine and hesperidin. But other four methods have less change ofthe extraction rate(less than 9.5%) than alcohol- precipitation and have more reduction of the contents of ephedrine and hesperidin(about 7%-10% and 10%-18%). To Xuanfeiqingre Granules, the sequence of clarifying is alcohol-precipitation > ZTC1+1 clarifying agent > JH clarifying agent > chitosans > Ultracentrifugation. 6 ) The optimum prepartion conditions for inclusion were established as: oil: β -CD was 1:6, the inclusion temperature and time were at 70 ℃ and for 1h, the rate of stirring for inclusion was 250r/min.Conclusion: 1) According to the experiment, alcohol-precipitation is also the best clarifying method. 2) The verification test show that the prepartion conditions for inclusion of oil is reasonable and practical.
