As a kind of excellent sand-fixing plant, Artemisia sphaerocephala Krasch(ASK) extensively exists in the west of Inner Mongolia, Ningxia, GanSu and the south of Mongolia. Each part of ASK is very useful, the branches and leaves of which can be used as diet, and fruits of which is very practical for medicinal purposes. In one word, ASK itself is very valued for natural resources and environment. So It is very important to study chemical composition, structure and bio-activity of polysaccharides from ASK.Two kinds of acidic heteropolysaccharides ASPl and ASP3 were obtained from ASK seeds through hot water extraction, deproteinatation of enzyme and Seveg, and grading precipitation with ethanol. The compositions and chemical structures of ASPl and ASP3 were determined by acid hydrolysis, Smith degradation, IR, HPLC, 1HNMR and 13CNMR. Results showed that ASPl, with Mn=2.84X 105, was composed of Xy1 which is the main constituent, Glu and Ara monosaccharides. ASP3, with Mn=3.50X 105, was composed of Glu, Gal, Ara and Man in molar ratio of 2.1: 1.3: 1.0:1.4. curonic acid was existed in both ASPl and ASP3. Glucoside bonds of ASPl and ASP3 were mainly jointed by 1-3 and 1-4joint respectively. Both ASP1 and ASP3 own a and B configurations.To observe the effect of CASP and ASP on serum lipid profile in rats, rats were randomly divided into four groups, namely control, positive control (Simvastatin), crude artemisia sphaerocephala krasch seed polysaccharide (CASP) and artemisia sphaerocephala krasch seed polysaccharide (ASP). Results showed that CASP and ASP can reduce serum TC in rats and effect of ASP is better than that of Simvastatin, and CASP and ASP remarkably can reduce serum TG in rats (P<0.05). Effects of ASP 1 and ASP3 on clotting time of mice were evaluated by Capillary Method. Result showed that ASP1 and ASP3 can remarkably prolong the clotting time of mice (P<0.01).In this thesis, a lot of methods in chemistry, biology and pharmacology were used to study the structures and pharmacological activities of polysaccharides from ASK. The whole works are basis and pre-works for further exploiting and utilizing ofASK.
