| With different methods, three kinds of functional factors including immune protein factor, gibberellin and cyclic hydroxamic acid have been extracted from wall-broken cole bee pollen.The procedure of purification included immersion time 72h, ammonium sulphate precipitation, Ultrafiltration, Ion-exchange chromatography on DEAE SephadexA-25, SephadexG-50 gel filtration, FPLC Superdex G-75, etc. a immune factor composed of three polypeptide chains, as molecular weight of 9000 dal, was isolated and purified. The samples was purified and used for the study of immune function promoting on Low immunity model mice by cyclophosphamide, the immune experiment shows the protein has immune activity function promoting, the immune factor can increase significantly cellular immune and humoral immune function of immunosuppressed mice in High dosage induced (0.645g/kg) .The wall-broken Cole bee pollen lOOg is extracted by 1:10 of anhydrous ethanol , continuously stirred in darkness for two hours. And then by means of centrifuge, cleaning, vacuum concentration, adjusting pH value, anion exchange resin chromatography, propanone extraction,being blown to dry and freeze-dried, we use HPLC to detect the content of the GA derivative of the freeze-dried powder. The result shows that the content of theGA derivative is about 51.29%.The wall-broken cole bee pollen 100g is extracted by 1:10 of 14.5%acetonitrile (pH=2.0) for 14 hours under the normal temperature. AfterSephadexG-10 or G-25 and G-10 gel filtration, we collected the mainingradient of anti-BPH, DIBOA. The content of it is up to 0.015%(150μg/g) .In addition, the application of functional factors of bee pollen is also investingated and expatiated in this paper. |
