The transition-metal complexes is one kind of important nucleic acid probe and has been developed to be one of the important research field in resent years, it has been used to study aberrance of chromosome,examine tumour and virus, and used as fluorescent probe to detect the structure of DNA. So it's application is focal prospective.However, an amount of study about transition-metal complexes focus ruthenium compleses . we try to use anther transition-metal replace ruthenium , synthesize new fluorescent probes.This dissertation cosists of six chapters. In chapter 1,the significance of study about complexes band to nucleic acid isdescribed . the common sense about nucleic acid were also introduced, the history ,general concept, application in biochemistry, determine method were reviewed in detail.In chapter 2, the synthesize methods of octahedron transition-metal complexes have been studied .the synthetic route of [Ru( bpy)2Cl2] has been improved. Reaction conditions for some intermediates were investigaged, as a result, some intermediates total yieleds were increased. Ten new transition-metal mixed ligands complexes were synthesized and the IR above these complexes has been studied.In chapter 3, The binding of the complexes of the complexes to calf thymus DNA was investigated by using fluorescent spectrum and ultravioler visible absorption spectroscopy. The result indicate that when complexes were binded to ct DNA , fluorescent spectrum were enhanced and UV were weakened one of these complexes[ Ni ( phen) oPHPIP] has strong fluorescence when it was binded to DNA, so Ni ( phen) 2PHPIP is helpful to become a new kind of nucleic acid probe.In chapter 4,The electrochemical behavior of interaction of [Ni (phen) 2PHPIP]2+ with DNA was studied by cyclicvoltammetry .When DNA was added in the [Ni( phen)2PHPIP]2+ solution, both reduction and oxidation peak currents of [Ni ( phen ) 2PHPIP]2+ decreased with nochange in the peak potentials. The electrochemical parameters in the absent and present of DNA were estimated,. An electrochemical inactive supra molecular complex DNA-[Ni ( phen) 2PHPIP]2+ was formedoIn chapter 5, a new nucleic acid probe ,[Ni (phen) 2PHPIP]2+ ,was used to band to fs-DNA and ct-DNA. results showed that complexes can intercalate ct-DNA but can't intercalate fs-DNA .so this complex can be a fluorescent probe to detect the structure of DNA.In chapter 6, the research work of this thesis was summarized ,and the developing trend of the fluorescent probe was proposed.To sum up,in this thesis ,some new investigations and innovation have been carried out for the design an synthesis of some new nucleic acid probe. The properties of spectrum and banding DNA, the application of DNA structure detection, the mechanism of complexes band DNA have been studied in detail. Especially, the investigation of [Ni (. phen) 2PHPIP]2+ band DNA have been never find in the literature . A new theory and some valuable experiment date have veen provided for the investigation and exploitation nucleic acid fluorescent probe.
