The extract from calyxes and leaves of Roselle (Hibiscus sabdariffa, Linn) was previously reported as food pigment and biologically active components, but phytochemical or pharmacological properties of the saponins from the seeds of Roselle have not been reported to date. The optimum operating conditions for extraction of high quality saponins from Roselle seeds have been obtained with orthogonal design. The resulting product of extraction was subjected to chromatography on a silica gel column. TLC ( thin-layer chromatography) was carried out on silica gel and the spots of triterpene saponins were visualized by spraying with the Liebemann-Burchard reagent. The characters of oleanolic acid ( -amyrane) type isolated saponins were showed on basis of chemical and spectra methods. The two components, RS1 and RS2, were purified by repeated chromatographed on silica gel column. RS 1 and RS2 with 2 and 5 monosaccharides were characterized by IR, UV, HPLC,I3 C NMR and ESI+/MS spectrum, respectively. Surfactivity, bio-activity of saponin from Roselle were primarily determined. The reduced surface tension was showed and its dispersion force of lime soap was 2.5. The inhibition effect on mice with S180 ascitic cancer was displayed, the higer ILS was 35.29%.
