In order to solve the problem of dextran ?" cane-cooked-rice" (CCR) from sugarcane factories, a series of experiments were done to study on CCR. They included: 1. isolation and identification of the strain producing dextran; the result was that the strain belongs to leuoconostoc mesenteroieds, ,which can produce a harmful CCR from sucrose. 2. components of the fermentation product and its productive conditions; CCR is a sticky dextran composed of glucose, a kind of completely methylation ,waterless pyranoglucose;95% of its glucosidic band is a -(1,6); 5% is a -(1,3) and others, molecular weight is above several millions. Flask medium is : sucrose 6%, yeast extract 0.4%, (NH4)2HPO4 0.6%, KH2PO4 0.1%, MgSO4 0.1%, inorganic ion. Fermentive condition: inoculating quantity 10%, aerate concentration 50ml,pH7.0, temperature 25 癈, rotation speed lOOrpm, time 26h. 3. screening and identifying a high-productive strain which can produce a kind of enzyme to decompose CCR. With UV, Co80, LiCl mutagenesis,the highest prodution strain SB 126 was finally obtained; the process was : beginning strainDA12?((LiCl)SB126. Its enzyme activity increased 2 times, up to 70U/ml. 4. the optimum of flask fermentation was obtained and the characteristic of enzyme was studied, the suiTable. medium is: glucose 4%, (NH4)2HPO40.03%,KH2PO4 0.15%,MgSO4 0.1%- etc ; and the fermentive condition is aerate concentration 30ml,rotation speed 180rpm,pH7.0; temperature 30'C, inoculating quantity 8%,4d. At last the enzyme stability was studied .It is better when pH<5.0 and T<60癈.Ions of Zn2', Ca2', Co2' , Reactivate it , Mn2', Mgz-, Fe2' , Cu2'on the contrast. |
