This paper focused on the analyzing method on the activity of D- dehydrogenase, the relation between the activity of D-dehydrogenase and the production of D-ribose, D-ribose-producing bacterium抯 characteristics and its fermentation medium and conditions. The optimum conditions, under which the D-dehydrogenase were determinated, is p1-I 6.8, temperature 3700, and time 10 mintiues. According to the analyzing method of D-dehydrogenase, the positive taxis between the activity of D-dehydrogenase and the production of D-ribose was acquaired, that the production of D-ribose was determined by the activity of D-dehydrogenase.The D-ribose- producing bacterium was obtained according on its D-dehydrogenase. After original strain, Bacillus Pumilius JFO 1 (shi) is N~ ion implantation induced mutagenesis., D-ribose-producing bacterium 5- 3,being shikimic acid deficient and high D-dehydrogenase-activity, was selected . The mutant could accmumulated 6.0% in fermented broth. Characteristics of D-ribose-producing bacterium 5-3 were studied. Compared with original strain, some differences were listed as follows. It is shikimic acid deficient,lower ability of spore formation, higher activity of D-dehydrogenase and production of D-ribose. The optimum fermentation conditions of strainD-ribose by mutant 5-3 was studied, and defined the optimum medium components: glucose 18%, corn steep 2.6%, ammonia-sulfate 0.85%, glycitol 2%, and with this medium components, its production of D- ribose could accumulated 6.35%.
