Cyciopentadecanolide as one kind of the macrocyclic musk, which has strong and graceful animal musk fragrance that is exquisite and lasting, is broadly used in many fields, such as beverage, food, cosmetics, tobacco and etc.The process using 1,15-pentadecanedioic acid as starting material to synthesize cyclopetade-canolide is studied in this paper. Mono-methyl 1,15-Pentadecanedioic acid was synthesized through di-esterization and trans-esterification.15-hydroxypentadecanoic acid prepared for cyciopentadecanolide was obtained after selective reduction of ester base and carboxyl. This article focused on the synthesis process of 15-Hydroxypentadecanoic acid, as well as the recycled and reused of byproduct.A method of reversed phase high performance liquid chromatography (HPLC) with internal standard was applied to the quantitative analysis of 1,15-pentadecanedioic acid and 15-hydrox-ypentadecanoic acid, which was an important material in the synthesis of cyclopentadecanolide. Optimum conditions were determined as follows:column packed with C18(4.6×250mm), lauric acid as internal standard.The mobile phase consists of a mixture of metholanol, acetonitrile and water(80:5:15,0.1% trifluoroacetic acid) with a flow rate of 1.2 mL/min and UV detection performed at 210nm.
