With the development of bamboo industry and the increasingly grown-up transgenic techniques nowadays, using biotechnology to improve the gene of bamboo plants has been concerned widely. And the stable bamboo regeneration system is indispensable platform of transgenic breeding. This research focused on the callus induction, proliferation and differentiation of Bambusa multiplex var.riviereorum and the embryo callus induction of Bambusa multiplex. Otherwises, cytological observation was used on callus. The results were:1 The callus culture of Bambusa multiplex var.riviereorum(1) The callus induction of Bambusa multiplex var.riviereorum: The best explant type were germfree plantlets, and the suitable medium for callus induction were MS+2,4-D 3mg/L + 6-BA 0.5mg/L. It could promote callus generate on the condition of red, blue light and dark, and VitaminC100mg/L as browning-preventative reagent.(2) The callus proliferation of Bambusa multiplex var.riviereorum:The best medium for callus proliferation were MS+2,4-D5mg/L+NAA1mg/L+6-BA/L,and sucrose 40g/L as the best concentration. Hydrolyzed casein, L-glutamine and proline were harmful to callus proliferation, And the effect of L-cysteine was little.(3) The different types of callus: there were two types of callus in induction: Typeâ… :This type of callus was close,light yellow and light white,had irregular shape. Typeâ…¡:This type of callus was sticky, translucent and hygrophanous. Through cytological observation and analysis,two types of callus were embryogenic callus, Typeâ…¡was an unhealthy state in callus induction and proliferation of Bambusa multiplex var.riviereorum.2 The embryo callus induction of Bambusa multiplex(1) Disinfecting embryo with 0.1% mercuric chloride, and the time should be controlled between 20 min and 30 min.(2) Crosscutting was the best processing of embryo in callus induction.(3) The suitable medium for embryo callus induction were MS+2,4-D2mg/L+KT 1mg/L.(4) There were three types of callus in induction of Bambusa multiplex embryo: Typeâ… :This type of callus was light yellow and loose;Typeâ…¡: This type of callus was gray and dense,had rough surface;Typeâ…¢:This type of callus was translucent, sticky and hygrophanous.Through cytological observation and analysis, type I was non-embryogenic callus, type II and type III were embryogenic callus, and type III was an unhealthy decline state of type II.
