As a significant epigenetic modification, DNA methylation plays a critical role in regulating gene expression and maintaining genomic stability in higher animals and plants. In this study, DNA methylation profiles were investigated in self-compatible line and self-incompatible line of Brassica oleracea var. acephala. The methylation-sensitive amplified polymorphism (MSAP) technique was employed in assessing methylation regulated genes to establish the genomic methylation pattern and rate of kale during self-incompatibility state. The main results were summarized as following:1. MSAP (Methylation-Sensitive Amplification Polymorphism)analysis was conducted to elucidate the level and pattern of DNA methylation at CCGG sites in self-compatible line and self-incompatible line.The results showed that:the ratio of methylation of is from 26.19% to 29.02%,12.64% to 13.45% for lateral, and 13.44% to 15.57% for cytosine sites. So genomic DNA methylation of Brassica oleracea var. acephala was mainly occurred in inside cytosine sites, the average level is 14.13%. The highest level of cytosine methylation level of the four experiment materials is self-incompatible line '9'. Self-incompatibility line of '9' overall percentage of methylation than the self-incompatibility system in the' Gan 6 'higher than 2.34%. Self-compatible line and the self-incompatible line reciprocal cross the progeny genomic DNA methylation level of disparity.2. From the MSAP patterns of the four experimental materials, we selected many bands which are different at the same point of DNA sequence, most of them belong to flower development-related genes, structural genes, chaperones, transcription factors. We found the the gene S-12 SRK and SP11A, which had different MSAP methylation pattern from the four experiment materials, at the same time thay play a role in process of self-incompatibility.
