Bio-chip is a new technology with many characteristics such as high-speed,high-efficiency, high-throughput and so on, bio-chip provide a good technical platform for the detection of microbial genes. Surface plasmon resonance sensor is a refractive index analytical techniques. The specific reaction of biological molecules and probe will result in changes of refractive index, so analyze the SPR signal can achieve the following objectives:biological molecular recognition, molecular concentrations and kinetic parameters. SPR biosensor is fast,sensiti-ve,effective,real-time and without being labeled or purified,It is being used in a wide variety of areas such as proteomies,drugdiseovery,cliniealdiagnosis, foodanalysis,environmental monitoring and so on.In this research ,our aim is to establish a method for the fabrication of DNA chip and the analysis technique based on SPR technique. The first step, take the probes of saccharomycete and beta hemolytic streptococcus and their complementary sequences as subjects, self-assembled monolayer method was applied to immobilize probe on the surface of gold film and find the ways to optimize the conditions, The protocal for the fabrication of DNA chip and the SPR analysis technique was established. In the second step, Scanning Surface Plasmon Resonance (SPR) detection system was applied to research and analyze the hybridization information of DNA chip.In our research, we got achievement as below:1.The thiol probe was immobilized on the surface of gold film using self-assembled monolayer method,and succeed in hybridization. 2. Scanning Surface Plasmon Resonance (SPR) detection system was succeed in researching and analyzing the hybridization information of DNA chip.The results showed that the hybridization has a good specificity.3. Scanning Surface Plasmon Resonance (SPR) detection system has a good sensitivity and stability. SPR sensor chips have been developed in this paper,which not only greatly reduces the equipment running cost and improves the analytical capacity of the SPR biosensor,but also lays the foundation for further application of the SPR biosensor and development of domestic SPR biosensor.
